The thing a lot of you are missing about this comic is that Jo did a series of these comics in the past where the red hellbear looked just like the yellow one does now.
And yes, the yellow one paid off bounty to kill the red one so he could be enchanted instead. Enchantress is not amused.
Well, i don't play dota, but after the explanation by Kenpachi Ramasama and Ewig Custos, the comic became understandable... i gotta admit it's clever, even if i'm not a huge fan of the game

So the yellow monster, which is weaker and smaller, has a crush on Enchantress and is jealous of Red Monster. So Yellow monster hired Bounty Hunter, the Hero in the first panel, to kill the big red monster. That way, Enchantress has to charm Yellow Monster and spend time with him.
Okay so, I don't play DotA but I think the joke is:
Enchantress is usually played as jungler thanks to a spell that lets her mind control creeps/neutral monsters. The red monster in panel 2 is the larger of two enemies from a monster camp, and is frequently charmed because it's strong.
Eventually the next strip continues the joke like the usual few-comics long thingies

got to say, even as a dota 2 player, the joke does fly over your head...
its either BH purposely killing him to ruin enchantress or enchant already got the smaller one first... both of which arent really funny or anything.
I really dont think the former has even been used.

For those who are wondering:
Yellow hellbear is in love with Enchantress, but she always chooses the stronger red hellbear. So he put a bounty on his head so Enchantress would have no other choice but to pick him.
Still a better love story than Twilight.

The joke is, enchantress accidentally enchanted the small creep, and now hellbear is left alone to die by himself. Bounty Hunter excels at ganking people when no one is around.
